I'll start with what I thought was the most mind-blowing twist was that Chuck might actually be God. I'm basing this on his disappearing act and the whole him as the writer of their lives thing, and being a prophet as a cover.

I never did think getting Satan to jump in the cage was going to be that easy so I was expecting things to go pear-shaped for Sam when he said yes, which y'know, they did. And I'm glad they did because it set up for a fantastic final fight.

I enjoy the subtleness to the scenes which could've easily been long-winded speeches, prime example is last weeks episode where Death was a matter of fact figure rather than villiainous and manicial. So I greatly enjoyed the confrontation between Michael and Lucifer, it really showed the points of view these characters had and just how petty the whole thing was.

Also, assbutt.

After the initial shock of seeing Cass blown to meat-bits I figured Bobby would die too, especially after he shot at Lucifer, which didn't disturb me too much as I had a feeling they would both be back soon. I just had no idea they would be back so soon and that Cass would be full blown angel again.

Just a little side comment. Since Bobby died and was brought back, I'm wondering if this means Crowley still has ownership over his soul.

So Sam's back but to what extent? Is it really Sam, and if so what does that mean for the Winchester brothers.

Don't know if this is a spoiler or not, but better safe than sorry.Bobby's coming back for season 6! He said so on his twitter. Happy about this because it wouldn't be the same without him

My guess for Season 6 would maybe be Sam bringing Dean back to the hunting world, to contrast the first season.
It should start with Sam and Adam starting up hunting together, then the big bad for the season captures Adam (because he's Michael's vessel? Seems a logical reason...) Sam turns to Dean for help, and then they go off hunting together again.

EDIT:
And then at the end of every episode it can be Dean yelling at Sam for not telling him he was back. Because they need to have the same emotional talk at the end of every episode.

So after watching the episode again on saturday with the girlfriend something that kind of niggeled at me during my watch suddenly jump out and banged my head on the table the second time through.

When Dean rings Chuck and the holy prophet mistakes Dean for Mistress Magda, a phone sex worker, it struck me as odd that the camera lingered a little too long when Chuck covers the ad with his drink.

It wasn't until the second time watching the ep that the name was a pretty obvious allusion to Mary Magdalene, the female apostle to Jesus. Dean even alludes to this himself when he comments on Becky when he says, "..a whole virgin hooker thing going-on".

So, this brings me to my point. Is Chuck really Jesus, and not God? It would make sense that Jesus, the son of God, would have an arc angel protecting him on Earth. It's also a clever way to keep God out of the picture but still have a holy figure repersented to actually give a damn in the going-ons of the Apocolypse.
